biomarkers of toxicity

The term toxomiR refers to a selection of 19 microRNAs that are enriched in various tissues. Toxicant-induced changes in the transcription and secretion of toxomiRs are highly sensitive to toxic effects occuring in the liver, kidney, central nervous system, heart, muscle, lung, pancreas, vascular injury, and due to endocrine disruption. ToxomiRs can be analyzed in serum or plasma.

This review recently published, gives detailed insight into the clinical utility of circulating microRNAs as biomarkers of toxicity.

Analyses with the toxomiRTM panel are available as in-house service by TAmiRNA and can be used for:

  • non-invasive screening of toxic effects using serum or plasma
  • identifying and monitoring safety risks in drug development
  • early detection of tissue injury

Frequently Asked Questions (FAQ)

1. How many samples can we measure with one toxomiR™ panel?

Upon request the analysis can be targeted to a specific set of miRNAs/tissue.

2. Can we use rodent or other non-human samples to test toxicity?

The toxomiR™ panel is intended to be used with human samples. However, orthologs for toxomiRs do exist in other species, allowing the use of adapted version of the Panel in non-mammalian species. To assess the feasibility of such projects, please contact us.

3. Which quality controls are included in the assay?

5 control primers to monitor assay and sample quality are included in the panel.

5. I want to analyze a sample type that is not listed as tested yet, can I do that?

Please contact us to discuss the possibilities.

6. What is the recommended RNA extraction method?

TAmiRNA´s toxomiR™ panel has been standardized to a volume of 200 µL plasma as starting material to ensure high RNA yield and prevent inhibition of downstream PCR amplification using the miRCURY™ RNA isolation kit. For further information read our guidelines detailing RNA purification and sample preparation for the toxomiR™ kit.

7. What is the sensitivity of the panel?

LOD: 10 copies/µl (6 replicates, 95% positive detection); LLOQ: 35 copies/µl 8. What is the reproducibility of the panel? Intra-assay (n=6) ≤ 20%, Inter-assay (n=3) ≤ 20%
